BCR::ABL1 tyrosine kinase domain (TKD) variant detection moved to Next Generation Sequencing

As of the 17th October 2022, BCR::ABL1 tyrosine kinase domain (TKD) variant detection for the North Thames Genomic Laboratory Hub (GLH) will move to next-generation sequencing (NGS).

Testing will be provided by the SIHMDS Department within North-West London Pathology (NWLP), Imperial NHS Trust. This service will replace the current Sanger sequencing methodology offering an improved lower limit of detection (3% variant allele frequency (VAF)) to comply with recent expert recommendations.

Sample requirements

Please send 15-20ml peripheral blood or 3-5ml bone marrow in EDTA that will be receipted within the SIHMDS laboratory <72 hours from collection. In instances where this is not possible, please send >25ul of cDNA.

Test requests

Please use the NT-GLH Specialist HaemOnc request form and ensure the following information is provided:

  • Diagnosis (e.g. CML [M84.8], AML [M80.15], ALL [M91.11] or AL other [M89.17]).
  • BCR::ABL1 transcript type.
  • Current therapy.
  • Reason for referral.
  • Address for report.
  • Requesting clinician.
  • Patient demographics.

Please note that testing will only be performed on samples with a BCR::ABL1 IS ratio >0.1%.

The turnaround time for this test is 21 calendar days. Please note that this is currently awaiting full UKAS accreditation.
